Interbus will handle all your needs. I myself I would take the regular bus and save a couple of hundred.

Interbus charges per person, $57 each way per person SJ to Manuel Antonio. If you take your favorite ($114), it might be more than a private car. I know the issues with using Uber in CR, but its estimates are useful. Uber estimates are 40,500 – 56,850 Colones. About $70 - $100.

Interbus gets the job done. I’ve used it in the past. I place it somewhere between the Tico bus and private transportation. You spend an hour picking up a van full at various hotels. Then stop at the large souvenir shop on the way to Jaco where they shuffle people to a different van depending on destination. Hub and spoke model. Then spend another hour with hotel drop offs at destination. It can double the time of your trip. We spent over two hours at the souvenir shop once when a van was late due to bad roads.

If there were two people, I would definitely go private car over Interbus

Chances are your favorite knows how to use the regular bus if you want to go economy.

For transportation you can't beat Vanessa whose contact info is listed at the beginning of this thread, she only provides transportation services. If you're staying at SL (very good choice) you should have time to find a nice lady to join you or stop in Jaco before Manuel Antonio. Then there's always Skokka CR you can Google if you prefer making arrangements prior to arriving in SJ.

For 2 people and you want comfort it is a no brainer to get Vanessa to take you for about the same price as 2 shuttle bus tickets.
You can go real cheap and fairly comfortably by regular buses that run from a terminal on calle 5 and Ave 18 I think. You can easily find out where it is and go there and buy tickets for about $5 per person each way.
From Quepos you can buy bus tickets to Jaco and also Jaco to San Jose. Air conditioned buses, only hold on to your carry on stuff as it gets stolen if left in the overhead bins. 4 to 5 buses a day.

I used Vanessa to return to the airport from the Sportsmen's. She was there early for the pickup. An immaculate and comfortable Hyundai sedan. She speaks great English and is very interesting to talk to. Fare was $30.

Be advised I tried to use her service from the airport but it was a Wednesday and her license sticker prohibits driving downtown on Wednesday s.
