I am a Marlboro man, and I found Marlboros for aroyund two bucks per pack. However, these are "export" Marbs and may taste a bit different from U.S. Marlboro Reds. The local, cheaper brand closest in flavor to U.S. Marlboros is Belmont, about $1.50/pack.
Other brands, such as Winston or Camel are hard to find. Remember that Costa Rican Customs rules allow you to bring a carton from home.

Also, the Presidente, del Rey and Sportsmen's all offer laundry/valet service. I'm sure other hotels do also. The hotel valet/laundry is however, expensive.
3. Is American express Credit cards accepted at most places?
No. Your hotel, probably. Rental car agencies, yes. Big chain restaurants, maybe.
VISA is the credit card of choice here.
4. Is booze availible 24 Hrs. ?
By the drink at the Blue Marlin Bar nad the lobby bar at the Hotel del Rey, other nightclubs until the wee, wee hours. By the bottle: Most supermercados carry a full liquor selection, and fairly cheap.
Suggestion: Buy your favorite top-shelf American brands at the Duty Free shop as you come into Baggage Claim in the airport. Buy local rums, guaro, beers at a supermercado during daylight hours.
5. What are the main suites like at the Hotel Castillo like ie: amenties?
No idea, as I've never stayed there. I know that each room has a key-operated safe, and that the Castillo serves a crappy free breakfast, fine if you like hardboiled eggs.

Also, don't forget Happy Hour, most afternoons at around five o'clock in the Blue Marlin bar at the Hotel del Rey. Visit the Sportsmen's Lodge early evenings as well. You'll find somebody with whom to wing, no problema!
